چکیده

=> 7' 7 ,72 Recent studies of chronic hypercapnia in guinea pigs and rats demonstrated a biphasic shift in the blood oxygen dissociation curve, a diminution of heme-heme interaction and a significant decrease in the oxygen capacity of blood which was not due to decreased hemoglobin content. The latter finding suggested production of non-functional hemoglobin during hypercapnia. We measured the concentration of oxidized (met) hemoglobin and found an increase from control values of 2. 69% to 8. 73% at 1 day, 6.4% at 3 days and 6.2% at 7 days of 15% CO2 exposure. The values are correlated with the biphasic changes in oxygen affinity (P50) and heme-heme interaction (n) of blood oxygen dissociation curves during hypercapnia. Hemoglobin oxidation is enhanced by low pH, and inhibition of red cell catalase by CO2 resulting in increased oxidation by H2O2 may also be partially responsible for methemoglobin formation. The persistence of methemoglobin results from inhibition by low pH of the enzyme systems which convert methemoglobin to hemoglobin.

Interspecies variability in propylene glycol dinitrate-induced methemoglobin formation.

Interspecies variability of propylene glycol dinitrate (PGDN)-induced methemoglobin formation was studied in vitro employing erythrocytes from four separate species. The net rate of methemoglobin formation was significantly different among species with dog greater than guinea pig greater than rat greater than or equal to human. This order of susceptibility was maintained in stroma-free hemolysa...

Bulletin of Experimental Biology and Medicine, USSR. 47:50, 1959. A lethal dose of sodium nitrite was injected into rats. All died. The average methemoglobin content of blood collected just before death was 72 per cent. Another group of rats re...

Catalysis of soluble hemoglobin oxidation by free iron on sickle red cell membranes.

Abnormal deposition of hemichrome on the inner aspect of the sickle red cell membrane promotes premature cell demise. The steps proximate to hemichrome formation in these cells are poorly understood. To test the hypothesis that the pathologic deposits of free ferric iron located on the inner aspect of sickle cell membranes would be redox active and promote oxidation of soluble oxyhemoglobin, we...

Decreased CYP3A expression and activity in guinea pig models of diet-induced metabolic syndrome: is fatty liver infiltration involved?

BACKGROUND In humans, CYP3A drug-metabolizing enzyme subfamily is the most important. Numerous pathophysiological factors, such as diabetes and obesity, were shown to affect CYP3A activity. Often considered a precursor state for type II diabetes, metabolic syndrome exerts a modulating role on CYP3A, in our hypothesis. OBJECTIVE To evaluate the effect of metabolic syndrome on CYP3A drug-metabo...
